بررسی تنوع ژنتیکی ارقام گندم نان (Triticum aestivum L.) به کمک نشانگرهای ریزماهواره و تجزیه‌ارتباطی برای دو صفت پرولین و فروکتان تحت تنش سرمای بهاره

چکیده

در این تحقیق تنوع ژنتیک 22 رقم گندم نان با استفاده از نشانگرهای ریزماهواره و صفات فیزیولوژیک مرتبط با تحمل به تنش سرمای دیررس بهاره در شرایط کنترل‌شده بررسی گردید. به این منظور آزمایش فاکتوریل با دو عامل رقم در 22 سطح و دما در 4 سطح C˚8+ (شاهد)، C˚2+، C˚0 وC˚ 2- در قالب طرح بلوک‌های کامل تصادفی اجرا گردید. نتایج تجزیه واریانس حاصل از داده‌های فیزیولوژیک نشان داد که اثرمتقابل رقم در سرما در هر دو صفت در سطح احتمال خطای 1% معنی‌دار بود. بر اساس نتایج به‌دست آمده در صفت پرولین در تنش C˚ 2-، بالاترین میزان پرولین، متعلق به رقم مغان2 و کمترین مقدار مربوط به رقم گلستان بود. همچنین در صفت فروکتان در تنش شدید، (C˚2-)، بالاترین مقدار قند فروکتان متعلق به رقم اوحدی و کمترین مقدار مربوط به رقم دز بود که برای بررسی تنوع ژنتیکی در سطح مولکولی از 21 جفت آغازگر ریزماهواره استفاده شد که 11 جفت توانستند چندشکلی مطلوبی را نشان دهند و در مجموع 19 آلل شناسایی گردید. تعداد آلل‌های تولید شده با میانگین 27/2 آلل در هر مکان ژنی، از 2 تا 4 آلل متغیر بود. میانگین سطح اطلاعات چندشکلی (PIC) برابر 62/0 و از 09/0 تا 98/0 متغیر بود. نتایج حاصل از رگرسیون گام‌به‌گام ارتباط معنی‌دار 2 نشانگر واجد اطلاعات را به‌ترتیب با عناوین Xgwm174 و مرتبط با صفت پرولین در شرایط تنش شدید و Xgwm642 را با صفت فروکتان در شرایط شاهد نشان داد.

چکیده [English]

In this study, the genetic diversity of 22 bread wheat cultivars was investigated through microsatellite markers and physiological traits under controlled condition. For this purpose an experiment was implemented via those varieties with four treated levels of cold stress (8 (control), +2, 0, -2 Celsius) in factorial arrangement in a randomized completely block design (RCBD). Results of variance analysis showed the effects of interaction between cultivars spring cold levels is signification at the 1% probability of error level. According the results of trait Proline content in -2ºC, The highest level of this trait belongs to Moqane2 and the Golestan was the lowest one. Fructan content, Also in this level of cold stress, showed Ohadi and Dez cultivars, respectively. To investigate the genetic diversity at the molecular level were used from 21 microsatellite primer pairs. The 11 pairs of them could show good polymorphism and totally identified 19 alleles. The mean number of produced alleles was 2.27 alleles per marker locus and the range of alleles at loci was 1 to 4. The polymorphic information contents (PIC) values of the loci had ranged from 0.09 t0 0.98 with average 0.62. Results of stepwise regression, shows the significant relationship 2 markers, Xgwm174 Proline-related traits in intense stress condition and Xgwm642 with fructan-related In terms of control.
