بررسی اثر تنش خشکی بر مولفه های جوانه زنی وفعالیت آنزیم های آنتی اکسیدانت در ژنوتیپ های لوبیا(vulgaris L. Phasseolus )

چکیده

تنش‌ خشکی، یکی از مهمترین تنش‌های محیطی است که تاثیر منفی در تمام مراحل رشد و نمو گیاه، به‌ خصوص مرحله جوانه‌زنی بذر می‌گذارد. به منظور بررسی اثر تنش خشکی بر جوانه‌زنی بذور ژنوتیپ‌های مختلف لوبیا، 40 ژنوتیپ لوبیای معمولی در ‌چهار سطح تیماری تنش خشکی ( شاهد، 4- ، 8- و 12- بار پلی اتیلن گلیکول 6000) به صورت آزمایش فاکتوریل در‌ قالب طرح کامل تصادفی در سال 1393 با سه تکرار مورد بررسی قرارگرفت. نتایج نشان داد که تنش خشکی باعث کاهش معنی‌دار درصد و سرعت جوانه‌زنی، طول ریشه‌چه و ساقه‌چه، وزن تر و خشک ریشه‌چه و وزن تر و خشک ساقه‌چه می‌شود. به منظور مطالعه‌ آثار بیوشیمیایی مقاومت به تنش خشکی در مرحله جوانه‌زنی سه ژنوتیپ 18 ، 25 ، 28 حاصل از غربالگری تست جوانه-زنی و رقم تجاری یاس در چهار سطح تیماری تنش خشکی (شاهد، 2- ، 4- و 8 - ) مورد بررسی قرار گرفتند. آنزیم‌های مورد بررسی در مرحله جوانه‌زنی کاتالاز و گایاکول پراکسیداز بودند، که ژنوتیپ 1189 دارای بیشترین میزان فعالیت آنزیم کاتالاز و گایاکول پراکسیداز نسبت به شاهد خود بود، همچنین ژنوتیپ 916 دارای کمترین میزان فعالیت این آنزیم‌ها بود. با بررسی صفات مورد مطالعه و شاخص‌ها، ژنوتیپ 1189 با مد نظر قرار دادن خصوصیات فیزیولوژیکی و بیوشیمیایی بهتر در قیاس با سایر ژنوتیپ ها و رقم یاس به عنوان شاهد در مرحله جوانه‌زنی به تنش خشکی متحمل بوده و به عنوان بهترین ژنوتیپ در شرایط این پژوهش معرفی می‌شود.

چکیده [English]

Drought stress is one of the major causes of reducing crop improvement. To evaluate the effects of drought tolerance on seed characteristics the tolerant and sensitive seed genotypes of common bean were tested in petri dishes containing polyethylene glycol 6000 with concentration equal to 0, -4, -8 and -12 bar, according to CRD design with three replications in factorial arrangement. 9 day later, radice and caulice lengths were scored. The results showed that drought stress decreased rate and percentage of germination, radice and caulice lengths, fresh and dry weight of radices, and fresh and dry weight of caulice. To study biochemical processes in germination stages, genotypes 18, 25, and 29 and commercial variety of Yas were screened for polyethylene glycol 6000 with concentration equal to 0, -4, -8 and -12 bar. By evaluating of the studied traits and indices, with considering physiological and biochemical characteristics, it could be conclude that genotype 1189 was drought tolerant one in seedling stage in comparison to the rest of genotypes and Yas as a check variety.
