بررسی اثر حفاظت فراسرد به‌روش شیشه‏ای کردن بر شاخص‏های رشد و نمو جنین‏های زیگوتی برنج

چکیده

با توجه به اهمیت ژرم‏پلاسم ارقام برنج ایرانی و کاربرد آن در برنامه‏های اصلاحی و تبادل مواد ژنتیکی، اجرای تحقیقاتی در خصوص حفاظت این منابع ژنتیکی ارزشمند، ضروری است. در این زمینه، روش نوین حفاظت فراسرد که نگهداری نمونه‏های زیستی در ازت مایع با دمای 196- درجة سانتی‌گراد است، کاربرد دارد. ازاین‌رو در پژوهش حاضر، حفاظت فراسرد جنین‏های زیگوتی هفت رقم برنج بومی و اصلاح‏شده به‌روش شیشه‏ای‏کردن بررسی شده است. در این تحقیق، جنین‏ها به‌‏مدت 1، 7 و 30 روز در ازت مایع نگهداری شدند. پس از گذراندن دورة انجماد، جنین‏ها از ازت مایع خارج شده و به‌مدت 2 دقیقه در دمای 40 - 38 درجة سانتی‌گراد قرار داده شدند و سپس به محیط کشت MS انتقال یافتند. در بین ارقام مورد مطالعه برای صفت درصد جوانه‏زنی نهایی، تفاوت معنی‏داری وجود نداشت؛ اما در صفت شاخص بنیه در سطح 5 درصد و در سایر صفات در سطح 1 درصد تفاوت معنی‏دار وجود داشت. در اکثر صفات مورد بررسی، بین مدت زمان‏های نگهداری در ازت مایع تفاوت معنی‏داری وجود نداشت؛ ولی نسبت به زمان شاهد کاهش معنی‏دار بود. بررسی سایر روش‏ها و تیمارهای حفاظت فراسرد ژرم‏پلاسم ارقام برنج بومی ایران توصیه می‏شود.

Because of the importance of Iranian rice germplasm and its application in the rice breeding programs and international genetic resources exchanges, some researches are necessary to preserve these valuable genetic resources. For this purpose, the cryopreservation of the biological samples in liquid nitrogen at -196 °C can be used. In this research, the cryopreservation of the zygotic embryos of seven rice indigenous and modern cultivars was investigated by vitrification method. The embryos were stored in the liquid nitrogen for 1, 7 and 30 days. After cryopreservation, the embryos were transferred at 38-40 °C for 2 min and then were cultured on MS medium. There were no significant differences among the studied cultivars in the final germination percentage trait, but there were significant differences in vigor index trait in level %5 and in other studied traits in level %. In most traits, there was no significant difference among the storage times in liquid nitrogen while there was a significant decrease in the studied traits in comparison with the control. Investigation of the other cryopreservation methods and treatments on Iranian rice indigenous cultivars germplasm could be recommended
