بررسی تنوع ژنتیکی لاین‌های نخود با استفاده از نشانگرهای مولکولی ISSR

چکیده

در این بررسی برای مطالعۀ تنوع ژنتیکی 56 لاین نخود زراعی از نشانگر مولکولی ISSR استفاده شد. از 34 آغازگر ارزیابی‌شده برای تکثیر قطعات DNA ژنومی لاین‌های نخود، نه آغازگر با تولید 35 نوار چندشکل، الگوهای نواربندی مناسبی تولید کردند. میانگین درصد چندشکلی برای همة آغازگرها 01/66 درصد با دامنة تغییرات 25 تا 100 درصد بود. میانگین تنوع ژنتیکی نی و شاخص اطلاعات شانون در همة آغازگرها به‌ترتیب 325/0 و 492/0 به‌دست آمد. بر مبنای ضریب تشابه جاکارد، لاین‌های FLIP 01-40C و  FLIP 02-84دارای کمترین تشابه ژنتیکی (166/0) و لاین‌های FLIP 03-6C و FLIP 03-8C دارای بیشترین تشابه ژنتیکی (1) بودند. تجزیة خوشه‌ای به‌روش UPGMA و با استفاده از ماتریس شباهت جاکارد انجام گرفت. برش دندروگرام در فاصلة 51/0 لاین‌ها را در شش گروه مجزا قرار داد.گروه‌بندی کاملاً متمایز و فواصل شایان توجه لاین‌ها در گروه‌های مختلف در این دندروگرام، بیانگر تنوع مولکولی زیاد لاین‌های نخود مورد بررسی است. نمایش لاین‌ها در یک نمودار سه‌بعدی براساس سه مؤلفة اصلی اول حاصل تجزیه به مؤلفه‌های هماهنگ، تا حدی گروه‌بندی حاصل تجزیة خوشه‌ای را تأیید کرد و توانست لاین‌ها را از هم تفکیک کند.

چکیده [English]

In this research, ISSR technique was applied to assess genetic diversity among 56 chickpea lines. Out of 34 primers, nine primers produced appropriate banding patterns and produced 35 polymorphic bands in chickpea lines. Mean percentage of polymorphism of the primers was 66.01 with range of 25 to 100 percent. Mean of Nei's genetic diversity and Shannon information index over all primers were 0.325 and 0.492, respectively. Based on Jaccard's similarity coefficient, lines of FLIP01-40C and FLIP02-84 had the lowest similarity (0.166) and lines of FLIP03-6C and FLIP03-8C had the highest similarity coefficient Cluster analysis using UPGMA method and Jaccard's similarity matrix was done. The dendrogram was cut at 0.51 scaled interval and categorized the lines in six distinct groups. Distinct grouping and large genetic distances among the chickpea lines stated a good molecular variation. Showing the lines by a tri-plot based on the three first coordinates from PCoA, verified the clustering and efficiently separated them
