The results showed that PGPRs lead to membrane stability, root development, etc. To identify the potential miRNAs involved in drought stress, the publicly available EST sequences of the plant were obtained from NCBI and blasted against the previously known Plant miRNAs. Ultimately, five distinguished potential miRNAs were acquired in the plant (belonging to four families: miR408, miR5021, miR5565e and miR9559). Candidate miRNAs were involved in growth, stress tolerance, etc. miR9559-3P expression decreased in drought and bacterial inoculation conditions in drought sensitive and tolerant plants. However, in the absence of bacterial inoculation, the expression of miR9559-3P was higher in sensitive plants. miR9559-3P might increase sensitivity to drought stress by negatively regulating role and targeting a higher percentage of target genes in sensitive plants. However, the tolerant plant with a special regulatory display and reduced expression of miR9559-3P reduced the degradation of miR9559-3P transcripts, increased the expression of the target gene and improved stress tolerance. Since PGPR modulate miRNA expression and lead to drought tolerance and increase plant yield, using these bacteria as biofertilizer can be a suitable alternative method of chemical fertilizer.  }, keywords = {Drought stress,EST,miRNA,PGPR,Phaseolus vulgaris L}, title_fa = {شناسایی miRNA‌ها و ژن‌های هدف آن‌ها در Phaseolus vulgaris L. تحت تنش خشکی و PGPR}, abstract_fa = {باکتری­‌های محرک رشد گیاه (PGPR) با تعدیل بیان miRNA، بهبود ریشه و باعث تحمل خشکی می­شوند. جهت بررسی اثر توام miRNA و PGPR بر تحمل به خشکی لوبیای معمولی در یک آزمایش فاکتوریل کاملا تصادفی در سه تکرار، ژنوتیپ­های صدری و یاس در شرایط خشکی 30% و FC = 60% و PGPR (تلقیح با جدایه اول، دوم و مخلوط دو جدایه) به­همراه شاهد بررسی شدند. تنش خشکی پنج هفته پس از کاشت اعمال (به مدت 10 روز) شد و سپس از سه­ برگچه دوم و ریشه نمونه­گیری شد. نتایج نشان داد که PGPR­ها منجر به پایداری غشا، بهبود ریشه و... می­شوند. ­miRNA‌های دخیل در خشکی بر اساس همولوژی بین ­EST‌های NCBI و ­miRNA‌هایmiRBase  باBLASTn  شناسایی شدند. ­miRNA‌ها متعلق به خانواده‌­های miR408، miR5021، miR5565e و  miR9559هستند و در رشد، تحمل به تنش و ... نقش دارند. مشاهده شد که میزان بیان miR9559-3P در شرایط خشکی و تلقیح باکتری در گیاه حساس و متحمل به خشکی کاهش یافت. در حالت عدم تلقیح باکتری، میزان بیان miR9559-3P  در گیاه حساس بیشتر بود. احتمالا در گیاه حساس، miR9559-3P با نقش تنظیم کنندگی منفی و هدف قرار دادن درصد بیشتری از ژن­های هدف، باعث افزایش حساسیت به تنش شد، اما گیاه متحمل با یک نمایش تنظیمی خاص و کاهش بیان  miR9559-3باعث کاهش تخریب رونوشت­‌هایmiR9559-3P، بیان بیشتر ژن هدف و بهبود تحمل به تنش شد. با توجه به این­که PGPR­ها با تعدیل بیانmiRNA  منجر به تحمل خشکی و افزایش تولید گیاه می­‌شوند، استفاده از آن‌­ها به­‌عنوان کود زیستی یک روش جایگزین مناسب کود شیمیایی می­باشد.}, keywords_fa = {تنش خشکی,EST,miRNA,PGPR,Phaseolus vulgaris L}, url = {https://ijfcs.ut.ac.ir/article_86725.html}, eprint = {https://ijfcs.ut.ac.ir/article_86725_bf4c3d842e1b68098bb523067c44fadc.pdf} }
