Salt and drought tolerance and high seedling vigor of rice is one of the important traits in the establishment of seedling in direct planting system and saline and drought environments. In this research, the mapping population of F2:4 families derived from the cross between local variety Gharib (salt and drought tolerant) and improved variety Sepidroud (salt and drought sensitive) was used for identification of molecular markers linked to salt and drought tolerant genes at Faculty of Agriculture and Natural Resources, Gonbad Kavous University and Biotechnology Laboratory, Guilan University in 2010-2012.  A linkage map of 2475.7 cM with an average interval of 10.48cM was constructed using 105 AFLP markers and 131 SSR markers. QTL analysis using composite interval mapping detected ten QTLs for germination stress index, vigour seed index, traits germination rate, germination percentage, radicle length, plumule length, coleorhiz length, plumule dry weight, coleoptile fresh weight and seedling dry weight under drought stress conditions of either mannitol (8) and salinity (8 dS). Among the identified QTLs, seven QTLs associated with drought stress, and seven QTLs associated with salt stress, explained over 15% of the total phenotypic variance of traits. It seems that these QTLs can be the major QTLs controlling traits of drought and salinity tolerance and with more confidence they could be used in marker assisted selection program.}, keywords = {drought and salinity,germination stress index,rice (Oryza sativa L.),seed germination}, title_fa = {مکان‌یابی QTLهای کنترلکنندۀ تحمل به تنش‌های خشکی و شوری در مرحلۀ جوانه‌زنی برنج}, abstract_fa = {خشکی و شوری، دو تنش غیرزیستی مهم در تولید برنج در سراسر جهان محسوب می­شوند. تحمل به شوری و خشکی و قوی­تر بودن اجزای جوانه در برنج، از مهم­ترین صفات در استقرار بهتر گیاهچه در خاک­های شور و خشک است. این پژوهش در سال‌های 1390-1388 در دانشکدۀ کشاورزی و منابع طبیعی دانشگاه گنبد کاووس و آزمایشگاه بیوتکنولوژی دانشکدۀ علوم کشاورزی دانشگاه گیلان، به‌منظور شناسایی نشانگر­های پیوسته با ژن­های تحمل به شوری و خشکی، از جمعیت F2:4 حاصل از تلاقی رقم بومی غریب (متحمل به شوری و خشکی) و رقم اصلاح‌شدۀ سپیدرود (حساس به شوری و خشکی) انجام گرفت. نقشۀ پیوستگی حاصل از 105 نشانگرAFLP  و 131 نشانگر ریز­ماهواره، 7/2475 سانتی‌مورگان از ژنوم برنج را با فاصلۀ متوسط 48/10 سانتی­مورگان بین دو نشانگر مجاور پوشش داد. تجزیۀ QTL با استفاده از روش مکان‌یابی فاصله­ای مرکب، در مجموع برای شاخص تنش جوانه­زنی، شاخص قدرت بذر، صفات سرعت جوانه­زنی، درصد جوانه­زنی، طول ریشه­چه، طول ساقه­چه، طول کلئوریز، وزن خشک ساقه­چه، وزن تر کلئوپتیل و وزن خشک گیاهچه تحت شرایط تنش خشکی حاصل از مانیتول (8- بار) و تنش شوری (8 دسی­زیمنس) تعداد ده QTL مشترک را شناسایی کرد. از بین QTLهای شناسایی‌شده، هفت QTL مرتبط با خشکی و هفت QTL مرتبط با شوری، بیش از 15 درصد تغییرات فنوتیپی مربوط به صفات را توجیه کردند. به‌نظر می­رسد که بتوان این QTL­ها را در زمرۀ QTLهای مهم کنترل‌کنندۀ صفات تحمل به خشکی و شوری محسوب نمود و در صورت تأیید از آنها در برنامۀ گزینش به‌کمک نشانگر استفاده کرد.}, keywords_fa = {برنج (Oryza sativa L.),تنش خشکی و شوری,جوانه‌زنی,شاخص تنش جوانه-زنی}, url = {https://ijfcs.ut.ac.ir/article_50340.html}, eprint = {https://ijfcs.ut.ac.ir/article_50340_c68132ae4bfa2e359902aeb5af255585.pdf} }
